Plan a worksheet run: total determinations including blanks, duplicates and spikes, reagent volume with dead volume, and how many 96-position plates the run fills.
Try sample values — fills the form with the worked example below.

Total determinations
–
Reagent needed (mL)
–
96-position plates
Equation: Total determinations = samples × replicates + blanks + duplicates + spikes. Reagent (mL) = determinations × reagent per determination + dead volume. Plates = total determinations ÷ 96, rounded up to the next whole plate.
Method: Worksheet planning per ISO/IEC 17025:2017, clause 7.2 (selection and verification of methods) and ISO 7218 (quality controls in microbiological examinations): blanks, duplicates and spikes are built into the run, not added afterwards.
Assumptions: every determination uses the same reagent volume; the dead volume covers pipetting and priming losses only.
Limitations: the plate count is a layout aid — it does not check that controls sit in valid plate positions, and it does not replace the method worksheet itself.
Rounding: determinations and plates are whole numbers; reagent to 1 decimal place; intermediates unrounded.
This is a calculation aid, not a laboratory report — confirm against your accredited method before citing a value. Records are kept only in this browser on this device (localStorage); this is a device-only record aid, not a shared multi-user LIMS, and nothing is sent to any server.

Worked example: 12 samples × 2 replicates, plus 2 blanks, 2 duplicates and 2 spikes, 5 mL per determination and 10 mL dead volume: 30 determinations, 160.0 mL of reagent, 1 plate.

Check the reagent stock against the volume needed before starting the run — a shortfall mid-run wastes the whole worksheet. Tool 266 of 500 — GIFSQ
