Work out the enzyme for a batch from the required activity per kilogram of substrate and the activity stated on the enzyme preparation.

Try sample values — fills the form with the worked example below.

–
Total activity needed (U)
–
Enzyme needed (g)
–
Enzyme per kg substrate (g/kg)
Equation: Total activity (U) = dose (U/kg) × substrate (kg). Enzyme (g) = total activity ÷ activity (U/g). Enzyme per kg = enzyme (g) ÷ substrate (kg).
Method: Enzyme dosing by activity units: the dose and the activity both come from the supplier data sheet and the application note, in the units of the assay used (for example FCC units). One unit is defined by the assay — units from different assays are not interchangeable.
Assumptions: activity is the value for the preparation as supplied, at the stated assay conditions; the dose applies to the substrate weight, not the whole batch.
Limitations: activity falls with heat, pH outside the working range and storage time — the calculated dose is a starting point for a trial, not a guarantee.
Rounding: activity to 1 decimal place; enzyme weight to 2 decimal places; intermediates unrounded.
This is a calculation aid, not a laboratory report — confirm against your accredited method before citing a value.
Worked example: 100 kg of substrate at 500 U/kg with an enzyme of 10,000 U/g: total activity = 50,000.0 U; enzyme = 50,000 ÷ 10,000 = 5.00 g (0.05 g/kg).

Enter values above to see results.

Activity units are assay-specific: never mix units from different assays. Use the dose and activity from the same supplier note. Tool 212 of 500 — GIFSQ
