Allergen Testing Methods
“Is this product really allergen-free?” Testing answers it — if you pick the right method for the question. ELISA, PCR, and lateral flow each detect allergens differently, and each has blind spots. Matching method to purpose is the whole skill.
The methods compared
- ELISA: detects allergen proteins via antibodies — quantitative, the standard for product testing and cleaning validation. Blind spot: heavily processed proteins may evade detection.
- PCR: detects allergen DNA — useful where protein methods fail (highly processed foods), but DNA presence doesn’t always track protein allergenicity.
- Lateral flow: rapid on-site strips — qualitative yes/no for surfaces and quick product checks. Less sensitive; screening only.
- LC-MS/MS: the confirmatory reference — identifies specific allergen peptides; used for disputes and method validation.
Choosing the method
- Cleaning validation: ELISA swabs (quantitative) or lateral flow (rapid) — prove the clean worked.
- Product verification: ELISA on finished product — quantify against your risk assessment.
- Processed foods: consider PCR or LC-MS/MS where heat/processing may have altered proteins.
- Always: validate the kit for your matrix — chocolate and spices humble many kits.
Why it matters
Allergen testing is what makes allergen management verifiable instead of faith-based. But no single method sees everything — the protein changed by your process might be invisible to the antibody but still allergenic. The professional approach layers methods: rapid tests for immediate decisions, ELISA for routine verification, and mass spec when the stakes (or the matrix) demand certainty. And document the validation — “we use Brand X kit” isn’t a method justification.
Match the test to the question: screening with ELISA or lateral flow, confirmation with PCR or LC-MS/MS — and validate for your matrix before trusting any of them.
