SOP: Salmonella Detection in Foods

Salmonella testing is non-negotiable for many food categories, and regulators expect the full enrichment-to-confirmation workflow. Skipping the pre-enrichment step is the classic mistake that lets injured cells slip through. This SOP follows the internationally recognized culture method.

Microbiological testing protects consumers from pathogens and tracks hygiene across the plant. Because living organisms are the analyte, aseptic technique and precise incubation conditions are not optional extras; they are the measurement itself. A single lapse in sterility or temperature control invalidates the result.

Purpose

To detect Salmonella species in foods by selective enrichment and plating with biochemical and serological confirmation, reporting presence or absence in the analytical unit.

Scope

Applies to raw meats, poultry, dairy, produce, spices, and environmental samples. Standard analytical unit is 25 g.

Equipment and Materials

  • Buffered peptone water (BPW) for pre-enrichment
  • Rappaport-Vassiliadis (RV) broth and tetrathionate (TT) broth for selective enrichment
  • Xylose lysine deoxycholate (XLD) agar and a second selective agar (for example Hektoen or brilliant green)
  • Triple sugar iron (TSI) agar and lysine iron agar (LIA) slants
  • Salmonella antisera (polyvalent O and H) or validated rapid confirmation
  • Incubators at 35 to 37 C and 41.5 C
  • Stomacher, sterile bags, pipettes

Procedure

  1. Aseptically weigh 25 g of sample into a sterile bag. Add 225 mL of BPW and stomach 2 minutes.
  2. Incubate the pre-enrichment at 35 to 37 C for 18 to 24 hours. This resuscitates injured cells.
  3. Transfer 0.1 mL to 10 mL of RV broth (incubate at 41.5 C) and 1 mL to 10 mL of TT broth (incubate at 35 to 37 C). Incubate both 24 hours.
  4. Streak each enrichment onto XLD agar and the second selective agar. Incubate at 35 to 37 C for 24 hours.
  5. On XLD, suspect colonies are red with black centers. Pick at least 5 suspect colonies per plate.
  6. Inoculate TSI and LIA slants. Typical Salmonella: TSI alkaline slant with acid butt, H2S positive, gas variable; LIA purple throughout (lysine decarboxylase positive).
  7. Confirm typical isolates by serology (agglutination with polyvalent antisera) or a validated alternative method.
  8. Report as detected or not detected in 25 g.

Recording Results

Report “Salmonella detected in 25 g” or “Salmonella not detected in 25 g” with the method reference. Record all media lots, incubation times and temperatures, and confirmation results.

Quality Control

Verify each new lot of culture media with reference strains before use: the target organism must show typical growth and morphology, and a non-target strain must be inhibited or show atypical growth as expected. Include a positive control (reference strain) and a negative control (sterile diluent) with every test batch. Incubate an uninoculated plate from each poured batch as a sterility check. Monitor incubator temperatures twice daily with a calibrated thermometer; do not use incubators found outside tolerance until corrected and re-verified. Track contamination rates in blanks; a rising blank contamination rate signals a systemic problem requiring investigation. Participate in proficiency testing at least annually and investigate any unsatisfactory scores with documented corrective action.

Troubleshooting

  • Overgrowth by competitors on plates: selective enrichment temperature drifted. Verify the 41.5 C incubator calibration.
  • No suspect colonies from a known positive: pre-enrichment too short or selective media expired. Check media QC with control strains.
  • Atypical biochemistry: some serovars are H2S negative. Confirm by serology rather than discarding.
  • False positives on XLD: Proteus and Citrobacter mimic Salmonella. The TSI/LIA and serology steps resolve them.

Safety Notes

  • Salmonella is a Biosafety Level 2 pathogen. Work in a biological safety cabinet.
  • Autoclave all enrichments and plates at 121 C for 30 minutes before disposal.
  • Report confirmed positives per local regulatory requirements and retain isolates as required.
